Hi,
As it takes 6 months for the fingernail to grow from base to tip and 12 months for the toenail to do likewise, it will take 6 months for loceryl to work for fingernails and 12 months for toenails.
Early clinical improvement (the base of the affected nail will begin to look normal again) may be seen after 3 months but loceryl application must be continued for the entire period to complete the treatment.

As Dr Paul has explained, the appearance of a brown line on your nail, known as melanonchia, is due to melanin within the nail. It's most often due to non-worrying causes. These include: Moles Freckles Fungal infection Pregnancy Trauma/injury (eg. from nail-biting) Bleeding under the nail Melanonchia due to moles commonly affect the thumbs, index fingers and big toes, like in your case. Of course, as you may have read, the internet will say that the big C word is a possible cause too (melanoma, a particularly aggressive type of skin cancer).
Sounds like you may have early acne issues, it would be good to seek consultation at a doctor who is interested about acne treatment. You will need a detailed consultation, examination and most likely Salicyclic Acid Peel in the clinic, and take home cleansers, topical lotions and serums to treat the issue completely.
